  1. 1. Multiple-Emitter Super-resolution Imaging using the Alternating Descent Conditional Gradient Method

    Kandidat-uppsats, Lunds universitet/Beräkningsbiologi och biologisk fysik - Genomgår omorganisation; Lunds universitet/Institutionen för astronomi och teoretisk fysik - Genomgår omorganisation

    Författare :Dolev Illouz; [2023]
    Nyckelord :super-resolution; alternating descent conditional gradient method; ADCG; multiple-emitter localization; nanoscale imaging; single molecule tracking; Physics and Astronomy;

    Sammanfattning : This thesis examines the state-of-the-art 2D super-resolution technique alternating descent conditional gradient (ADCG) method's ability to accurately localize fluorophores in diffraction-limited single molecule images (SMI) and analyze the impact of pre-processing and post-processing modules on ADCG's fluorophore localization. A synthetic dataset obtained from the 2013 Grand Challenge localization microscopy and a temporally linked dataset obtained from an unpublished set of Optical DNA mapping experiments performed by Jonathan Jeffet at the NanoBioPhotonix Lab at Tel-Aviv University were initially segmented to extract their noise parameters. 2. 2. Developing Automated Cell Segmentation Models Intended for MERFISH Analysis of the Cardiac Tissue by Deploying Supervised Machine Learning Algorithms

    Master-uppsats, KTH/Kemi

    Författare :Julia Rune; [2023]
    Nyckelord :Cell Segmentation; Cellpose; Supervised Machine Learning; MERFISH; Heart Failure; Cellsegmentering; Cellpose; Övervakad Maskininlärning; MERFISH; Hjärtsvikt;

    Sammanfattning : Följande studie behandlar utvecklandet av automatiserade cellsegmenteringsmodeller med avsikt att identifiera gränser mellan celler i hjärtvävnad. Syftet är att möjliggöra analys av data genererad från multiplexed error-robust in situ hybridization (MERFISH). LÄS MER

  3. 3. Single-molecule Imaging of the Cell Division Ring in Escherichia coli Using the ALFA-tag

    Master-uppsats, KTH/Tillämpad fysik

    Författare :Emma Westlund; [2023]
    Nyckelord :ALFA-tag; Escherichia coli; Divisome protein FtsZ; mEos3.2; Nanobodies; Nanotags; PALM; Plasmid transformation; sfGFP; Single-molecule Imaging; ALFA-tag; Escherichia coli; Divisomprotein FtsZ; mEos3.2; Nanokroppar; Nanotaggar; PALM; Plasmidtransformation; sfGFP; Enmolekyl-mikroskopi;

    Sammanfattning : The use of super-resolution (SR) microscopy is an important tool for understanding the inside mechanisms of bacterial cells. However, for SR imaging, the labelling of the proteins of interest is a great challenge as flourescent proteins (FPs) are often too big to be directly fused to the target protein and traditional immunolabelling with antibodies creates too long separation between the fluorophore and the target protein. 4. 4. Improving the temporal resolution of a microspectrometer for the study of the photophysics of enhanced green fluorescent protein

    Master-uppsats, KTH/Tillämpad fysik

    Författare :Lukas Rane; [2021]
    Nyckelord :Biophysics; EGFP; Fluorescence; triplet state; Biofysik; EGFP; fluorescens; triplet tillstånd;

    Sammanfattning : The use of fluorescent proteins as fluorescent markers has exploded over the last decades. In particular due to the development of advanced microscopy for live cell measurements, dynamic molecular studies down to single molecule levels and for superresolution microscopy. LÄS MER

  5. 5. A new approach to detect membrane proteins : Loading of marker molecules into liposomes for detection of single membrane proteins

    Uppsats för yrkesexamina på avancerad nivå, Uppsala universitet/Analytisk kemi

    Författare :Ida Styffe; [2017]
    Nyckelord :Liposomes; Membrane proteins;

    Sammanfattning : Membrane proteins are essential to the function of biological systems. The ability to map the placement and abundance of these proteins can facilitate several improvements within both medicine and biology. LÄS MER
