  1. 1. Towards time-resolved cryo-EM of SARS-CoV-2 replication-transcription complex and Staphylococcus aureus DNA gyrase

    Master-uppsats, Uppsala universitet/Institutionen för biologisk grundutbildning

    Författare :Anna Králová; [2023]
    Nyckelord :time-resolved cryo-EM; DNA gyrase; SARS-CoV-2; replication-transcription complex; protein purification;

    Sammanfattning : Time-resolved cryo-EM has already provided ground-breaking discoveries in various fields, including structural biology, biochemistry, and drug development. Compared to traditional structural biology methods where mostly stabilized conformations are reconstructed, the main advantage of time-resolved cryo-EM is its ability to capture dynamic processes in biological samples at near-atomic resolution, which allows for studying biological structures as they change and interact in real-time. 2. 2. Design, expression and purification of virus-like particles derived from metagenomic studies : Virus-like Particles (VLP) of novel Partitiviridae species, Hubei.PLV 11, and novel Soutern pygmy squid flavilike virus were designed, expressed using the bac-to-bac expression system and then pruified using various methods

    Master-uppsats, Uppsala universitet/Institutionen för biologisk grundutbildning

    Författare :Diyar Ayranci; [2021]
    Nyckelord :Virus; virus-like particles; VLP. Western Blot; SDS-PAGE; Chromatography; partitivrus; partitiviridae; flaviviridae; flavivirus; Hubei partitilike virus; Southern Pygmy Squid flavilike virus; protein purification; protein characterization; PCR; Cloning; metagenomic; strucutral biology; cryo-em; bac-to-bac expression system; vaccines; arbovirus; arthropod-borne virus;

    Sammanfattning : Viruses are entities which are made of a few genes and are reliant on obligate parasitism to propagate. Due to the obligate connection to their hosts, virus evolution is constrained to the type of host. Viruses however do transmit to evolutionary distinct hosts; in these cases, the phylogenetic relationship of the hosts usually are close. LÄS MER

  3. 3. The future of viral vectors for gene therapy

    Kandidat-uppsats, Uppsala universitet/Institutionen för biologisk grundutbildning

    Författare :Elias Ekstedt; Inna Fryckstedt; Hanna Hyllander; Josefin Jonsson; Elin Ring; Felix Wærn; [2021]
    Nyckelord :Gene therapy; adenovirus; alpharetrovirus; baculovirus; adeno-associated virus; lentivirus; herpes simplex virus; foamy virus; gammaretrovirus;

    Sammanfattning : Gene therapy is a fast growing technology that offers treatments for genetic diseases. The method is based on introducing genetic material into a patient to replace the disease-causing gene, using a vector. LÄS MER

  4. 4. Goodbye E.coli : Alternativa endotoxinfria produktionssystem till Escherichia coli

    Kandidat-uppsats, Uppsala universitet/Institutionen för biologisk grundutbildning

    Författare :Jenny Eriksson; Sofia Fasth; Cecilia Orrenius; Plavsic Milica; Linnéa Yuan Andersson; Marcus Westholm; [2020]
    Nyckelord :;

    Sammanfattning : Affibody is interested in replacing Escherichia coli as the production system for Affibody®-molecules in order to reduce the amount of resources needed for the purification step. The aim of this report is to present relevant endotoxin-free alternatives that would suit production. LÄS MER

  5. 5. Cloning, Expression and Purification of a Mutant Tomato Thymidine Kinase 1 for Crystallography

    Master-uppsats, Lunds universitet/Tillämpad biokemi

    Författare :Mohamad Nesrini; [2015]
    Nyckelord :protein expression; protein engineering; Azidothymidine; thymidine kinase; deoxyribonucleoside kinases; nucleosides; nucleoside analog; suicide gene therapy; tillämpad biokemi; applied biochemistry; BEVS.; Technology and Engineering; Chemistry;

    Sammanfattning : Thymidine kinase 1 from tomato (ToTK1) is a newly discovered deoxyribonucleoside kinase (dNK) that has been subject to study due to its in vitro and in vivo properties making it an interesting suicide gene candidate for the treatment of brain tumors. The ToTK1 phosphorylates the nucleoside analogue 3-azido-2,3-dideoxythymidine (azidothymidine, AZT) equally well as its natural substrate thymidine (dThd). LÄS MER
