  1. 1. Exploring alternative surfactants to replace PEG within lipid nanoparticles

    Master-uppsats, Lunds universitet/Livsmedelsteknik och nutrition (master); Lunds universitet/Bioteknik (CI); Lunds universitet/Bioteknik (master)

    Författare :Elina Edidovica; [2024]
    Nyckelord :Lipid nanoparticle; polyethelyne glycol PEG ; alpha-tomatine; N-hexadecyl beta d-maltoside HDM ; DOPC; DPPC; cholesterol; film hydration; stability study; DLS; Cryo-TEM; SAXS; pharmaceutical formulation; Chemistry;

    Sammanfattning : Over the past decade an increasing amount of studies has demonstrated that PEGylation not only precipitates reductions in drug delivery efficiency but also triggers the immune system to produce antibodies, specifically anti-PEG antibodies. The objective of my thesis is to explore alternative surfactants that could viably replace PEG within lipid nanoparticles. LÄS MER

  2. 2. Characterization of Mitilysin Pores by Cryo-electron Microscopy

    Master-uppsats, Uppsala universitet/Institutionen för biologisk grundutbildning

    Författare :Vladimir Novakovic; [2023]
    Nyckelord :Mitilysin; Mly; Structural Biology; Cryo-EM; pore; toxin; pore forming proteins; pore forming toxins; PFP; PFT;

    Sammanfattning : Pore forming toxins (PFTs) are a large group of proteins found mainly in bacteria with some exceptions found in animals. They bind and form pores in their target membranes and form pores, which leads to cell death. Among these are cholesterol-dependent cytolysins (CDC), which require the presence of cholesterol to bind target membranes. LÄS MER

  3. 3. Method development of magnetic cell isolation and DNA extraction of small cell populations from Ficoll-separated hematopoietic cells

    Kandidat-uppsats, Uppsala universitet/Institutionen för medicinsk cellbiologi

    Författare :Dominika Debowska; [2023]
    Nyckelord :Clonal haematopoiesis of indeterminate potential CHIP ; hematopoietic stem cell; automated cell separation; silica-membrane-based nucleic acid purification.;

    Sammanfattning : Clonal haematopoiesis of indeterminate potential, or CHIP are a family of mutations present in the general population. CHIP-mutations are prevalent in the haematopoietic stem cells and in the more mature cell populations, T-lymphocytes, B-lymphocytes and myeloid cells (CD3+, CD19+ and CD33+ cells) in blood. LÄS MER

  4. 4. A Novel Method for Finding Overlap Depth : Development of ArtiaX, a ChimeraX plugin

    Master-uppsats, Linköpings universitet/Tillämpad matematik; Linköpings universitet/Tekniska fakulteten

    Författare :Gunnar Arctaedius; [2023]
    Nyckelord :Cryo-electron tomography; image processing; surface visualization; overlap measurement; normal projection method; Kryoelektrontomografi; bildbehandling; ytvisualisering; överlapsmätning; normalprojektionsmetoden;

    Sammanfattning : Visualization and image filtering are important parts of cryo-electron tomography analysis. ArtiaX, a plugin developed for UCSF ChimeraX, has been extended to improve the functionality of these two parts. For the visualization, a method of moving 3D surfaces to remove overlap between them has been developed and implemented. LÄS MER

  5. 5. Towards time-resolved cryo-EM of SARS-CoV-2 replication-transcription complex and Staphylococcus aureus DNA gyrase

    Master-uppsats, Uppsala universitet/Institutionen för biologisk grundutbildning

    Författare :Anna Králová; [2023]
    Nyckelord :time-resolved cryo-EM; DNA gyrase; SARS-CoV-2; replication-transcription complex; protein purification;

    Sammanfattning : Time-resolved cryo-EM has already provided ground-breaking discoveries in various fields, including structural biology, biochemistry, and drug development. Compared to traditional structural biology methods where mostly stabilized conformations are reconstructed, the main advantage of time-resolved cryo-EM is its ability to capture dynamic processes in biological samples at near-atomic resolution, which allows for studying biological structures as they change and interact in real-time.
